Do you mean supporting MySQL as a mechanism for storing wiki pages,
or the ability to interface with MySQL for other purposes?
PmWiki defaults to storing its pages as flat files instead of a database,
although it's possible to write a MySQL backend for PmWiki. However,
we haven't encountered a strong reason for doing this -- see
http://www.pmwiki.org/wiki/PmWiki/FlatFileAdvantages for reasons why.
Interfacing with MySQL can be done via cookbook recipes -- see
http://www.pmwiki.org/wiki/Cookbook/MyPmWiki for an example.
